Oral isoindolinone GSPT1 glue degrader LG-NE-29 targets glioblastoma models

  • A viability-based phenotypic screen flagged GSPT1 as a vulnerability in glioblastoma cells, where resistance to temozolomide arises quickly.
  • Optimizing an isoindolinone series for metabolic stability gave LG-NE-29, a molecular glue that drives CRBN-dependent ubiquitination and proteasomal degradation of GSPT1.
  • It is reported to avoid the metabolic weaknesses of earlier scaffolds, and to be orally bioavailable with activity in glioblastoma xenograft models.
💡 Points

Isoindolinone CRBN glues usually die on metabolism, so fixing that liability is the real story here. Before I believe it, I'd want the GSPT1 DC50/Dmax, proteome-wide selectivity (IKZF1/3, other neosubstrates), and oral PK with brain exposure. GSPT1 degradation is also broadly cytotoxic, so the therapeutic window in normal cells matters.
